高通量发光细菌法在水质评价中的应用研究

摘要:
通过改进传统的单管式发光细菌毒性测试方法,测定了发光细菌悬液在高通量测试体系中的发光动力学数据和发光强度的校正因子,以评价发光细菌在高通量测试体系中的稳定性。采集了长三角示范区水乡客厅及周边区域12个采样点环境水样,并对该方法在评价环境水质中的适用性进行验证。结果显示,在75 min内,稀释10~100倍的发光细菌悬液的发光强度约为105~106RLU/s,各稀释梯度发光细菌悬液发光强度的校正因子平均值范围为0.78~1.10。各采样点环境水样的毒性单位范围为0.007~0.11,水质毒性分级均为I类,水质良好。研究结果表明,高通量发光细菌法稳定可靠,适用性强,可快速高效地评价环境水质。

Application of High-throughput Luminescent Bacterial Method in Water Quality Assessment

Abstract:
The luminescent kinetics data and the luminescent intensity correction factor of luminescent bacteria suspension in the high-throughput test system were measured to evaluate the stability of luminescent bacteria in the high-throughput test system by optimization of the traditional toxicity test method of luminescent bacteria in tubes. Environmental water samples were collected from 12 sampling sites in Shuixiangketing and its surrounding areas in the Yangtze River Delta demonstration area to validate the applicability of the method in assessing environmental water quality. The results showed that the luminescence intensity of the luminescent bacterial suspension diluted 10 to 100 times was about 105~106 RLU/s within 75 min, and the average correction factor of the luminescence intensity of each dilution gradient ranged from 0.78 to 1.10. The toxic units of environmental water in each sampling site ranged from 0.007 to 0.11, and the toxicity of water quality was grade I, indicating that the water quality was good. The results show that the high-throughput luminescent bacterial method is stable, reliable, and has well applicability, which can evaluate environmental water quality quickly and efficiently.
